Understanding and Applying Thioredoxin Systems in Plants

Research in the Buchanan Laboratory focuses on thioredoxin--a regulatory protein found in all types of living cell. Plants contain different types of thioredoxin, distributed throughout the plant.



We study the role of thioredoxin and related proteins in two areas.

   

I. Studies on regulation of enzymes of chloroplast stroma and thylakoid lum

We study regulation of chloroplast enzymes, emphasizing both the stroma and thylakoid lumen. This research is carried out in collaboration with Sheng Luan.

Recent research uncovered a previously unrecognized role of thioredoxin in regulating cysteine biosynthesis in association with a cyclophilin when plants are exposed to oxidative stress. In this as in other links to enzymes, thioredoxin acts as an eye for the chloroplast allowing biochemical processes of the organelle to adjust and make optimal use of available resources.



We have also uncovered a paradigm for redox regulation in photosynthesis in the thylakoid lumen. In contrast to the stroma, where the activation of enzymes is achieved by a reduction of disulfide groups conversion of (S-S to SH), activation in the lumen occurs in concert with oxygen evolution by the oxidation of sulhydryl groups (conversion of SH to S-S). The latter mechanism shows promise in the commercial production of disulfide-rich value-added proteins.




   

II. Improvement in the quality and yield of cereals

We have ongoing collaborations with several colleagues on projects to improve the quality and yield of cereals: Peggy G. Lemaux (UC Berkeley); Jun Yin and his group (Henan Agricultural University, Zhengzhous, China); and William J. Hurkman and William H. Vensel (USDA Laboratory, Albany, CA).

The grain harvested from the fields shown below displays improved properties. For barley, accelerated germination. For wheat, decreased allergenicity.





Studies with Dr. Salekdeh and his collaborators in Iran have uncovered a role for thioredoxin and redox in enabling wheat to survive drought conditions.



Recent studies in our laboratory have demonstrated that amylplasts of sink tissues contain a complete ferredoxin/thioredoxin system that regulates fundamental processes in the organelle. In this case, the signal is not light, but the increase in NADPH that results form the breakdown of increased sucrose arriving from the phloem when leaves are illuminated.



In effect, thioreodoxin of amyloplasts acts as a second eye in regulating processes of that plastid.



The amyloplast thioredoxin system complements the role of extraplastidic thioredoxin, that among other functions, acts as a signal in seed germination and seedling development.



In short, as established for chloroplasts, thioredoxin plays a fundamental role in a growing number of processes in heterotrophic plant tissues.
